The hosts, whose local FA has been under investigation over corruption allegations, did not play well and were punished after about an hour when Liverpool forward Luis Diaz assisted Daniel Munoz who found the ball superbly and into Alex Remiro's net.
The Spaniards have one more friendly match ahead - against Brazil on Tuesday, and on the same day the Colombians face Bulgaria's northern neighbor Romania.
In another fixture, Netherlands crushed Scotland 4-0. Tijani Reinders, Georginio Wijnaldum, Wout Weghorst and Donyell Malen scored for the "Tulips".
The world champion Argentina, as expected, prevailed over El Salvador with 3:0. The Albiceleste were without their big star Lionel Messi, but they still found success thanks to goals from Cristian Romero, Enzo Fernandez and Giovanni Lo Celso.
